Instructions for Submission of Outside Samples to the Columbia University Protein Core

Gel Bands or spots for MALDI or LC-MS/MS analysis:

1.  Stain the gel using one of the recommended stains on our web site at http://cpmcnet.columbia.edu/dept/protein/ or another MS-compatible stain. Best results are obtained with Coomassie-stained gels.

2.  While wearing gloves, cut the gel spot or band with a clean blade and put it in an Eppendorf tube. Be sure the cap is closed securely. No additional liquid is necessary. The samples may be stored at –20 until ready to send.

Samples in solution for mass determination: Send a 10-20µl aliquot in an Eppendorf or screw-cap tube. Make sure it is sealed securely. Tubes may be sent in Dry Ice or, if the protein is stable, at room temperature.

Samples for Edman (N-terminal) sequencing: For PVDF bands, cut with a clean blade and place in a tube. Send them at room temperature overnight. For samples in solution, follow the instructions above for mass determination samples.
